I have a lookup table in which the combination of several columns forms a key to lookup for a unique row.

Can I use a document cache to build such a lookup table?
How should the index the lookup table be defined?

Sure you can use the document cache for such a lookup table.
The Index for a Document Cache can include multiple keys. Simply add keys to the Index. You can then subsequently do a lookup based on that compound Index.
